    Raistonia solanacearum,the bacterial wilt pathogen, has a wide host range and genetic variability.Rapid and sensitive molecular techniques need to be developed for eariy detection and strain differentiation of the pathogen.Molecular techniques such as PCR and DNA hybridization have been succesfully used to detect and identify bacterial plant pathogens including R.solanacearum.These techniques were adopted under Indonesian condition, using purified and crude DNA from infected plant samples.An R.solanacearum specific DNA primer (OH/Y2) was used in the PCR test,and a DNA probe 5a67 were used in the non-radioactive hybridization test.The PCR techniqe could be used to detect R.solanacearum from infected plant samples in less than 5 hours.The DNA hybridization technique was applicable to differentiate strains ofR.solanacearum into three groups based on their DNA profiles

    Tujuan dari penelitian ini untuk mengembangkan media pembelajaran berupa buletin dalam bentuk buku saku untuk pembelajaran Fisika kelas VIII pada materi Gaya ditinjau dari aspek materi, konstruk, dan bahasa serta minat baca siswa. Penelitian ini termasuk penelitian pengembangan yang menggunakan metode Research and Development (R&D). Penelitian ini menggunakan model pengembangan model prosedural yaitu model yang bersifat deskriptif yang menunjukkan tahapan-tahapan yang harus diikuti untuk menghasilkan produk berupa media pembelajaran.Jenis data yang diperoleh bersifat kualitatif dan kuantitatif yaitu angket dan wawancara. Teknik analisis data yang digunakan adalah analisis deskriptif kualitatif dan kuantitatif. Hasil penelitian menunjukkan bahwa media pembelajaran yang dikembangkan berupa buletin Fisika dalam bentuk buku saku memiliki kriteria sangat baik berdasarkan penilaian dari ahli materi, ahli bahasa Indonesia, dan ahli media memberikan rata-rata penilaian sebesar 86,56%. Media pembelajaran yang dikembangkan juga memiliki kriteria sangat baik bila ditinjau dari peningkatan minat baca siswa. Hal ini terbukti pada hasil angket minat baca awal dan akhir yang diberikan kepada siswa yang memberikan rata-rata peningkatan sebesar 11,13%. Selain itu juga dianalisis dengan menggunakan uji-t berpasangan terhadap data masing-masing kelompok uji coba untuk mengetahui signifikansi dari peningkatan minat baca siswa. Untuk uji coba perorangan diperoleh hasil perhitungan thitung = 6,957 > ttabel = 1,943 dan nilai Sig. = 0,001 < 0,05 yang berarti sangat signifikan. Untuk kelompok kecil didapatkan hasil perhitungan bahwa thitung = 7,848 > ttabel = 1,725 dan nilai Sig. = 0,000 < 0,05 yang berarti sangat signifikan. Untuk kelompok besar juga didapatkan hasil perhitungan bahwa thitung = 20,214 > ttabel = 1,725 dan nilai Sig. = 0,000 < 0,05 yang berarti sangat signifikan. Simpulan dari penelitian ini adalah media pembelajaran berupa buletin dalam bentuk buku saku memiliki kriteria sangat baik bila ditinjau dari aspek materi, konstruk, dan bahasa serta minat baca siswa

    Cluster-Like Headache Secondary to Anamnesis of Sphenoid Ridge Meningioma: A Case Report and Literature Review

    Cluster headache is generally considered to be a primary headache; secondary cluster-like headache is quite rare, while cluster-like headache secondary to meningioma is even rarer. Here, we describe an unusual case with cluster-like headache 2.5 years after sphenoid ridge meningioma surgery. The cluster-like headache and meningioma were on the same side, and even at the same position. Furthermore, the cluster-like headache lasted for 6 months. In addition, the patient did not respond well to conventional treatments for cluster headache, such as oxygen inhalation, carbamazepine, and tramadol. Brain magnetic resonance imaging demonstrated a softening lesion, glial hyperplasia, and localized thickening and enhancement of the dura in the left frontal-temporal lobe. However, positron-emission computed tomography showed reduced metabolism in the left frontal-temporal lobe. Although the possibility of a primary headache cannot be completely eliminated, the association between cluster-like headache and probable tumor recurrence or postoperative changes should be considered

    Evidence for e+e−→γχc1,2e^+e^-\to\gamma\chi_{c1, 2} at center-of-mass energies from 4.009 to 4.360 GeV

    Using data samples collected at center-of-mass energies of s\sqrt{s} = 4.009, 4.230, 4.260, and 4.360 GeV with the BESIII detector operating at the BEPCII collider, we perform a search for the process e+e−→γχcJe^+e^-\to\gamma\chi_{cJ} (J=0,1,2)(J = 0, 1, 2) and find evidence for e+e−→γχc1e^+e^-\to\gamma\chi_{c1} and e+e−→γχc2e^+e^-\to\gamma\chi_{c2} with statistical significances of 3.0σ\sigma and 3.4σ\sigma, respectively. The Born cross sections σB(e+e−→γχcJ)\sigma^{B}(e^+e^-\to\gamma\chi_{cJ}), as well as their upper limits at the 90% confidence level are determined at each center-of-mass energy.Comment: 8 pages, 7 figures, 3 table

    Salivary Gland NK Cells Are Phenotypically and Functionally Unique

    Natural killer (NK) cells and CD8+ T cells play vital roles in containing and eliminating systemic cytomegalovirus (CMV). However, CMV has a tropism for the salivary gland acinar epithelial cells and persists in this organ for several weeks after primary infection. Here we characterize a distinct NK cell population that resides in the salivary gland, uncommon to any described to date, expressing both mature and immature NK cell markers. Using RORγt reporter mice and nude mice, we also show that the salivary gland NK cells are not lymphoid tissue inducer NK-like cells and are not thymic derived. During the course of murine cytomegalovirus (MCMV) infection, we found that salivary gland NK cells detect the infection and acquire activation markers, but have limited capacity to produce IFN-γ and degranulate. Salivary gland NK cell effector functions are not regulated by iNKT or Treg cells, which are mostly absent in the salivary gland. Additionally, we demonstrate that peripheral NK cells are not recruited to this organ even after the systemic infection has been controlled. Altogether, these results indicate that viral persistence and latency in the salivary glands may be due in part to the presence of unfit NK cells and the lack of recruitment of peripheral NK cells

    Artemisinin-Naphthoquine versus Artemether-Lumefantrine for Uncomplicated Malaria in Papua New Guinean Children: An Open-Label Randomized Trial

Artemisinin combination therapies (ACTs) with broad efficacy are needed where multiple Plasmodium species are transmitted, especially in children, who bear the brunt of infection in endemic areas. In Papua New Guinea (PNG), artemether-lumefantrine is the first-line treatment for uncomplicated malaria, but it has limited efficacy against P. vivax. Artemisinin-naphthoquine should have greater activity in vivax malaria because the elimination of naphthoquine is slower than that of lumefantrine. In this study, the efficacy, tolerability, and safety of these ACTs were assessed in PNG children aged 0.5–5 y.An open-label, randomized, parallel-group trial of artemether-lumefantrine (six doses over 3 d) and artemisinin-naphthoquine (three daily doses) was conducted between 28 March 2011 and 22 April 2013. Parasitologic outcomes were assessed without knowledge of treatment allocation. Primary endpoints were the 42-d P. falciparum PCR-corrected adequate clinical and parasitologic response (ACPR) and the P. vivax PCR-uncorrected 42-d ACPR. Non-inferiority and superiority designs were used for falciparum and vivax malaria, respectively. Because the artemisinin-naphthoquine regimen involved three doses rather than the manufacturer-specified single dose, the first 188 children underwent detailed safety monitoring. Of 2,542 febrile children screened, 267 were randomized, and 186 with falciparum and 47 with vivax malaria completed the 42-d follow-up. Both ACTs were safe and well tolerated. P. falciparum ACPRs were 97.8% and 100.0% in artemether-lumefantrine and artemisinin-naphthoquine-treated patients, respectively (difference 2.2% [95% CI -3.0% to 8.4%] versus -5.0% non-inferiority margin, p?=?0.24), and P. vivax ACPRs were 30.0% and 100.0%, respectively (difference 70.0% [95% CI 40.9%–87.2%], p<0.001). Limitations included the exclusion of 11% of randomized patients with sub-threshold parasitemias on confirmatory microscopy and direct observation of only morning artemether-lumefantrine dosing.Artemisinin-naphthoquine is non-inferior to artemether-lumefantrine in PNG children with falciparum malaria but has greater efficacy against vivax malaria, findings with implications in similar geo-epidemiologic settings within and beyond Oceania.Australian New Zealand Clinical Trials Registry ACTRN12610000913077.Please see later in the article for the Editors' Summary
